Enrichment of cross-linked species resulting from reaction of α-crystallin with DTSSP using 5 M GnHCl as the solvent for size exclusion chromatography. A: TSK3000 size exclusion HPLC chromatograms of denatured α-crystallin cross-linked by DTSSP (red trace) and denatured native α-crystallin not exposed to cross-linker (black trace). B: Coomassie stained 10% SDS-PAGE gel of four dialyzed contiguous fractions (F1-F4) eluting from TSK3000. The increase in the amount of crosslinked material as shown in F2, F3, and F4, relative to the results shown in Figure 1 was due to oxidation during preparation of the different fractions (results not shown). Based on SDS-PAGE, fraction 2 (F2), denoted by dashed red lines in A and eluting between 4.5 and 6.5 min (4.5-6.5 mL) was selected for digestion with trypsin.
