Figure 7. Effect of FGF-2 on actin cytoskeleton organization and cell shape of corneal endothelium on Descemet's membrane

Isolated corneal endothelium-Descemet's membrane complex maintained in DMEM-10 was incubated with one of the following conditions; FGF-2, FGF-2 + neutralizing antibody to FGF-2, FGF-2 + LY294002, FGF-2 + Y27632, FGF-2 + LY294002 + Y27632, FGF-2 + neutralizing antibody to FGF-2 + Y27632, or FGF-2 + LY294002 + Y27632 + neutralizing antibody to FGF-2 for 4 days. The concentrations used were 10 ng/ml for FGF-2, 20 μM for LY294002, 10 μM for Y27632, and 5 mg/ml neutralizing antibody to FGF-2. The tissues were fixed and stained with rhodamine-phalloidin (1:100) at 37 °C for 10 min. The data shown are representative of four experiments. Bar represents 10 μm.
