Figure 4. Retinoic acid (RA) concentration and time dependent regulation of RPE65 promoter activity
HEK293 cells were transfected with the 1345 bp RPE65 promoter-reporter construct and were changed into normal growth medium 24 h after transfection. Promoter activity was determined by luciferase assay. Activity (<10%) due to the effect of RA on vector alone was subtracted from all values. A: Incubating with RA (10-9 to 10-5 M) for 24 h. B: Incubation with 10-6 M of RA for 12, 24, or 48 h. Data are plotted as means; error bars represent the standard errors of the mean (n=3). Asterisks ("*") indicate statistical significance as compared with the control (*p<0.05; **p<0.01).
