A: Schematic presentation of the 5' region of the mammalian mimecan gene. The positions of transcription factor binding sites that are highly conserved between human and bovine mimecan promoters are indicated. Position of the E-box motif is conserved between human, bovine, mouse and rat promoters and its sequence shows 100% identity. The translation initiation start site (ATG) is indicated in human and bovine exon 2. Alternative splicing sites within the second exon of human and bovine mimecan genes are indicated with white lines. White rectangle within the arrow that marks the p53 binding site was used to mark the presence of half p53 binding sites in the bovine and mouse promoters. B: Schematic presentation of the human mimecan promoter/luciferase reporter constructs used in this study. The positions of transcription factor binding sites and primers used for the generation of promoter/reporter constructs are indicated.
