Figure 17. Coexpression of MP19To3red with Golgi-green protein in 293 cells

MP19To3redTO cells were cotransfected with the Golgi-pEGFP-N1 plasmid. This plasmid expresses green fluorescent protein fused to the N-terminal 81 amino acids of human β1,4-GT. This region of human β1,4-GT contains the membrane-anchoring signal peptide that targets the fusion protein to the trans-medial region of the Golgi apparatus. 48 h following transfection with Golgi-pEGFP-N1, the cells were induced with tetracycline and the cells were observed with confocal microscopy (EGFP filter set). The image was obtained using the inverted microscope with a 40x water objective.
