Figure 3. Effect of U73122 on cell proliferation and PLC enzyme activity
When CEC reached approximately 60% confluency, they were maintained in serum-free media for 24 h. Cells were then treated with U73122 in concentrations ranging from 1.0 to 40 mM in the presence of FGF-2 (10 ng/ml) for 24 h (A). Cell proliferation was assayed as described in the text. Control cells were maintained in the absence of FGF-2 and U73122. The inhibitory effect is presented as percent of the control. The inhibitory activity of U73122 on PLC enzyme activity was measured at concentrations ranging from 1.0 mM to 40 mM (B). After cells were labeled with myo-(2-3H) inositol in the inositol- and serum-free DMEM, they were treated with U73122 in the presence or absence of FGF-2 (10 ng/ml) for 1 h. Total inositol phosphate formation was assayed as described in the text. Data were presented as x-fold stimulation compared to the basal level obtained from the cells maintained in the absence of FGF-2 and U73122. Experiments were done in triplicate and repeated four times; data shown are from one experiment. Error bars represent the standard error of the mean.
