Phosphorylation of Cx46 in the lens cortex. Fiber cells of the lens cortex (3 lenses) were dissected, lysed, and homogenized, then Cx46 was immunoprecipitated and the samples were analyzed by SDS-PAGE and western blot as described in the text. A: Transferred proteins on western blots were probed with anti-phosphoserine (lane 1), anti-phosphotyrosine (lane 2), and anti-phosphothreonine antibodies (lane 3) all at 1:1000 dilution. B: As a control, the nitrocellulose membrane containing the transferred proteins were probed with anti Cx46 to detect the immunoprecipitated Cx46 and to determine equal load of Cx46 on the gel.
