A: A typical RNase protection analysis of IRBP gene expression showing the effect of actinomycin D on the forskolin-induced IRBP gene expression in laminin-pretreated WERI-Rb1 cells. Actinomycin D was added at the beginning of the time course of forskolin treatment. Treated, 3 day laminin-pretreated cells that were treated with forskolin (10 mM)/IBMX (0.5 mM) and actinomycin D (5 mg/ml); Control 1, cells that were neither treated with laminin/forskolin nor actinomycin D; Control 2, cells that were treated with DMSO and actinomycin D (5 mg/ml).
