Figure 1. A schematic diagram of the 2.4 kb Trc-Tox176 transgene construct used for microinjection

This construct consists of 277 bp 5'-fragment of GNAT2 (GNAT2 Pro), an attenuated diphtheria toxin-A subunit gene (Tox176), SV40 (intron and polyadenylation signal) and a 214 bp enhancer element from human IRBP (IRBP Enh). The preparation of the whole construct was described in Methods. The greater than sign marks the position of the 5' primer; the less than symbol marks the position of the 3' primer. These primers were used for screening transgene positive mice. MCS, multiple cloning site.
