Both MIP and the kanamycin resistance factor Neomycin phosphotransferase (NPTII) are flanked at their 3' end by Nopamine Synthase (NOS) flanking sequence , part of the wild-type A. tumefaciens Ti plasmid . Genes appended with this sequence have been demonstrated to be transferred to plant tissue more efficiently . Since MIP receives its own Cauliflower Mosaic Virus (CMV) promoter from the pSIN vector, it is does not matter in which direction it ligates into the final pVB6 vector.
