Figure 3. Subcellular localization of fusion proteins containing the SH3 domain of PLC-[gamma]1 (GST-SH3) and the (SH2)2-SH3 domain of PLC-[gamma]1 [GST-(SH2)2-SH3]
CEC were microinjected with either GST-SH3 (A and B) or GST-(SH2)2-SH3 (C and D). Two h after injection, cells were treated with FGF-2 (10 µg/ml) supplemented with heparin (10 µg/ml) for 4 additional h (B and D), while control cells were kept in the absence of serum (A and C). Cells were then fixed, permeabilized, and stained with anti-PLC-[gamma]1 antibody. Immunolocalization was determined as described in the text.
