For immunoblot analysis, the antigen of interest was dotted onto nitrocellulose. The membrane was incubated with 1 µg/ml anti-AGE antibody, and the reaction was visualized with indirect alkaline phosphatase immunochemistry as described in the Methods. A. Our anti-AGE antibody recognizes AGE-BSA, CML-BSA, and pentosidine-BSA, but not BSA, KLH, and matrix proteins. B. Immunoblot analysis of another polyclonal anti-AGE antibody [25] strongly recognizes CML-BSA, but not pentosidine-BSA. Significant reaction is seen against glycolaldehyde prepared AGE-BSA.
