The effects the iron chelator 2,2’dipyridyl-N,N-dimethylsemicarbazone on U-0126 and glucose oxidase induced increases in ferritin levels in retinal pigment epithelial cells and lens epithelial cells. Retinal pigment epithelial (RPE) cells and lens epithelial cells (LECs) were treated with 10 μM U-0126, 0.25 mU/ml glucose oxidase (GO), and/or 0.3 mM 2,2’dipyridyl-N,N-dimethylsemicarbazone (Dp) for 24 h. U-0126-treated cells were pretreated with the extracellular signal-related kinase 1/2 (ERK1/2) inhibitor for 10 min before GO and/or Dp was added. Ferritin levels were measured with enzyme-linked immunosorbent assay (ELISA). Each histogram bar represents the mean±SEM of at least six samples.
