Figure 1. The pFTM3GW lentiviral transfer vector. The pFTM3GW plasmid vector was constructed by adding restriction sites to the parent plasmid, pFTMGW. A SpeI site was added to the 5′ end of the hCMV enhancer, and a BstBI site was added to the 3′ end of the TB; both modifications duplicate existing sites on the opposite sides of the genetic elements, simplifying removal of the elements when desired. We also reordered the restriction sites in the multiple cloning site (MCS) to facilitate double-digestion reaction performance and subsequent cloning. ColE1 ori (bacterial origin of replication) and AmpR (ampicillin resistance gene) were used for plasmid replication. The enhanced green fluorescent protein (eGFP) is the reporter molecule. The cytomegalovirus promoter (CMV), long-terminal repeats (LTRs), splice donor and viral packaging sequences (SD/Psi), Rev Response element (RRE), central polypurine tract (CPPT) and Woodchuck hepatitis virus post-transcriptional regulatory element (WPRE) are viral elements.
