Figure 1. Expression of glucose-regulated protein 78 mRNA in human trabecular meshwork cells

A: Primary cultured human trabecular meshwork (HTM) cells were either exposed to 2.5 μg/ml Tm for 6 h or not treated. B: Primary cultured HTM cells were transiently transfected with the indicated expression plasmids. C: Primary cultured HTM cells were transiently transfected with the indicated expression plasmids, then cells were exposed to 2.5 μg/ml Tm for 6 h. Total RNA was isolated from each cell population, and subjected to RT-PCR for glucose-regulated protein78 (GRP78) mRNA (upper panels) or GAPDH mRNA (lower panels). The gel was representative of three separate experiments. The following terms were used: mock, cells transfected with pcDNA3.1(+) expression plasmid; wild, cells transfected with wild-type myocilin expression plasmid; and P370L, cells transfected with Pro370Leu mutant myocilin expression plasmid.
