A: Subconjunctival fibroblasts were treated with 10 nM phorbol 12-myristate 13-acetate (PMA) for the indicated times. Arrows mark the location where JNK proteins should appear. B: For inhibition experiments, we added bisindolylmaleimide I (BIS) or SP600125 to the serum-starved cells prior to PMA treatment. Expression and phosphorylation of c-jun N-terminal kinase (JNK) and c-jun were examined by Western blot analysis. We evaluated c-jun activation with an anti-phospho-c-jun specific antibody (Ser63 or Ser73). In the β-actin panel, it can be seen that equal amounts of proteins were loaded.
