Figure 1. PCR analysis of subtraction efficiency

To determine the efficiency of the suppression subtractive hybridization, measurement of secondary PCR product of the subtracted and unsubtracted human Müller cells cDNA has been performed. A: Control PCR product of the housekeeping gene β-actin. A decrease in the amount of amplicon present after subtraction can be observed when compared to unsubtracted. Samples were taken after 18, 23, 28, and 33 PCR cycles. B: Cellular retinaldehyde binding protein (CRALBP), a marker for Müller cells, is used as a control. A CRALBP amplicon is increased after subtraction. B represents before subtraction, A represents after subtraction.
