The PCR primers, S200 and A860, were added to first-strand cDNA from the neural retina or retinal pigment epithelium (RPE) of donor Y-005 (OD). Amplification was carried out to generate expected 609-bp and 495-bp fragments that correspond to the intact RGR and truncated RGR-d mRNAs, respectively. Parallel experiments were performed with (+) or without (-) reverse transcriptase. No PCR bands were detected in the controls. The about 600-bp (solid arrow) and about 500-bp (open arrow) PCR fragments from the RPE were sequenced and found to contain the 114-bp presence or deletion of exon 6, respectively.
