Triton X-100 extraction assay. Total protein extracts (lane 1) were subjected to extraction with either 1% Triton X-100 alone (lanes 2, 3), MCD alone (lanes 4 and 5), or combination of both (lanes 6 and 7) and probed with anti-Panx1 antibodies (upper panel); the same blot was reprobed with anti-Cx50 antibodies (lower panel). The asterisks mark the 58 kDa and 120 kDa Panx1 isoforms and the 50 kDa Cx50 that showed resistance to either Triton X-100 or MCD treatments but were partially extracted by combined MCD/Triton X-100 treatment. Even though all samples were equilibrated for the total protein and volume prior to extraction, protein recovery was reduced after treatments that included MCD. Lanes marked by "p" correspond to pelleted fraction, by "s" to supernatants, "+" to treated, and "-" to untreated fractions.
