Inhibition of EGFR activation reduces insulin-induced cell migration. A: THCE cells cultured in serum- and growth-factor-free medium were pretreated with aphidicolin and exposed to DMSO, 1 μmM AG1478, 100 nM wortmannin, or 50 μM GM6001 for 1 h. Confluent cells were scratched with a micropipette tip, and cell migration was determined after incubation with or without 5 μg/ml insulin for 12 h. Scale bar, 200 μm. B: Cells (2x10²) were seeded into 96-well plates and starved for 24 h. Cells were then grown under the indicated conditions and growth was assayed using the cell proliferation reagent WST-1 after 24 h. The absorbance values are given on the y axis.
