A pan-specific TGFβ antibody is unable to block the TGFβ-like effects induced by vitronectin or fibronectin substrata. A, B: Explants on vitronectin. C, D: Explants on fibronectin. E, F: Standard explants. G, H: Explants on laminin. B, D, F, H: Explants cultured with 50 μg/ml of TGFβ antibody. E: Explant cultured with 250 pg/ml TGFβ2. The presence of the antibody had no effect on cells migrating on vitronectin or fibronectin. The cells still showed irregular, migratory morphologies and lost their characteristic polarity and cobblestone-like packing arrangement (B, D). TGFβ2 induces characteristic phenotypic changes (E) but these are blocked by the presence of 50 μg/ml the TGFβ antibody (F). The presence of the TGFβ antibody alone had no effect on cells migrating on laminin. These cells maintained their polarity and cobblestone-like packing arrangement (H). The scale bar indicates 100 μm.
