Overexpression of K6W-ubiquitin delays the cell cycle in G2/M phase. HLEC were arrested in G0 phase by maintaining cells at confluence for 48 h and then infected with empty adenovirus or adenovirus encoding K6W-ubiquitin. After infection for 24 h, the cells were induced to resume the cell cycle by replating at about 20% confluence. The cells were then collected at the indicated times. The cells were fixed and stained with propidium iodide. DNA content was measured with a FACS Calibur Flow Cytometry System and the population of cells in each phase of the cell cycle was analyzed using ModFit LT software. Experiments were performed at least three times with reproducible results. The asterisk indicates a p<0.05 for the comparison of cells infected with adenovirus encoding K6W-ubiquitin to cells infected with empty adenovirus at the indicated times.
