Lys6 was replaced with Trp to create the dominant negative ubiquitin variant, K6W-ubiquitin (A). Wild-type and K6W-ubiquitin were expressed HLEC via an adenoviral vector and levels of endogenous ubiquitin conjugates in the cells were determined by western blotting using anti-ubiquitin antibody (B).
