Figure 5. Sensitivity of HIOMT gene transcription to different lots of serum
E7 retinal cells were cultured for three days in medium supplemented with different lots of fetal calf serum (FCS) or with newborn calf serum (NCS). Total RNA (15 μg per lane) extracted from approximately 10 million cells was analyzed on northern blot with the indicated 32P-labeled cDNA probes. HIOMT gene transcription was efficiently activated by one lot of FCS out of three, whereas red opsin gene transcription was activated by all lots of FCS. This variability was not observed for NCS, which consistently activated transcription of both HIOMT and red opsin.
