Figure 1. Conservation of the sequences flanking rhodopsin kinase transcription start sites

Alignment of sequences upstream of the human and mouse rhodopsin kinase (Rk) genes was performed using BestFit and BLAST algorithms. The scanned sequence included the entire genomic interval separating the first exons of adjacent, head to head-arranged Rk and H,K-ATPase β-subunit genes (human genomic clone XX-86039E12 on chromosome 13 (GenBank accession number BX537316) positions 10900-20239 and mouse genomic clone RP23-89F9 from chromosome 8 (GenBank accession number AC130818) positions 56370-64800). The entire area of identity by BestFit algorithm is shown and the regions of more rigorous sequence identity by BLAST algorithm are shown in red, blue, or green. The red sequence shows the conserved H1 sequence. The blue sequence marks the noncoding areas of significant BLAST identity (78%) and the green sequence marks the coding sequence (90+% identity). No other overlap was detected in the upstream noncoding sequences beyond those shown here. The hyphens above the human sequence demarcates the region included in the Rk-GFP transgene (-112 to +87). Positions are numbered arbitrarily relative to the most distal transcription start site for each gene. Bracketed numbers represent the nucleotide positions in the surveyed GenBank clones. The asterisk and plus sign represent transcription and translation start site region, respectively.
