Figure 2. Plasminogen and angiostatins in cornea conditioned medium

Human corneas were cut into 8 sections and each section was placed in a well of a 24 well plate and cultured for 3 or 27 h (A). In other experiments, the corneal sections were incubated in the presence or absence of 5 ng/ml IL-1β for 0, 2, 5, 8, or 24 h (B). Media samples were taken and separated by SDS-PAGE and electroblotted to nitrocellulose. The membranes were probed with two monoclonal antibodies specific for the K1-3 portion of the plasminogen (Plgn) molecule. A: Plasminogen and smaller molecular weight bands corresponding to K1-4 and K1-3 angiostatins were found in the supernatants from the organ cultures. B,C: IL-1β alters pattern of angiostatins at 5 h but not at 2, 8, or 24 h of culture.
