Figure 3. Oxidation of tryptophan in α-crystallin
Tryptophan (Trp) fluorescence (excitation at 290 nm, emission at 330 nm) and N-formylkynurenine fluorescence (excitation at 330 nm, emission at 440 nm) of normal and UV-C irradiated α-crystallin were measured using a Hitachi F-4500 spectrofluorometer. The relative fluorescence intensity of Trp was derived so that the intensity of non-irradiated α-crystallin was 100%. The relative fluorescence intensity of N-formylkynurenine was derived so that the intensity of UV-C irradiated α-crystallin at 50 J/cm2 was 100%.
