RNA was generated from the coding regions of PDE6α, β, γ and PDE-associated δ from Y79 cells and in vitro translated into protein. Reaction products were electrophoresed in an acrylamide gel and visualized by autoradiography. Arrows point to the full length protein that was obtained from each of the coding regions ruling out the possibility of a premature stop codon. To the right of the delta lane the upper arrow points to the 15 kDa delta protein, and the lower arrow points to the 11 kDa gamma protein. Based on the high degree of specificity of the PDE6 antibodies used, the additional faster migrating bands in the beta lane are likely minor degradation products; however, this was not proved to be true in this study. A size marker was run and the sizes of each product corresponded to the size of authentic PDE6 subunits (not shown). No product was observed in the absence of added template (not shown).
