Figure 5. Increase in emission in fluorescently labeled arrestin bound to R*P
AlexaFluor-594 labeled arrestins (10 μM) were mixed with sonicated phosphorylated rhodopsin (20 μM) in disc membranes and fluorescent emission measured in the receptor bound and unbound state. The fluorophore was excited at 340 nm, and emission measured at 5 nm intervals with a 100 msec dwell time. Fluorescence emission increased in the bound state for I72C and S251C.
