Figure 2. Nucleotide sequence and the PCR amplification of the βIGH3 region

A: Putative transcription factor-binding sites within 1 Kb of the 5'-flanking region of the βIGH3 gene, as determined by the Proscan, TFSEARCH, and MatInspector databases. The term SBE indicates putative Smad binding elements. The start codon (ATG) is the last three bases. The asterisk represents the transcription starting site at -65 bp position. B: The proximal regions of mouse and human βIGH3 promoter are highly conserved. Numbering is relative to the ATG start codon. C: Effects of DMSO on the synthesis of 1 Kb fragments. PCR products were analyzed on a 0.8% agarose gel. Lane 1 shows the PCR reaction without DMSO, showing no apparent band. Lane 2 shows the PCR reaction with 1% DMSO, showing a weak 1 Kb cDNA band. Lane 3 shows the PCR reaction with 5% DMSO, showing a strong 1 Kb cDNA band.
